چکیده

This indirect method for determination of desferrioxamine (DFO, deferoxamine, Desferal) and its chelated compounds ferrioxamine (FO) and aluminoxamine (AIO) in biological fluids involves metal chelate formation and solvent extraction, the metal being determined by Zeeman atomic absorption spectrometry (Zeeman-AAS). Special attention was paid to the pH and recovery of extraction, iron-aluminum interactions, extraction times and volumes, centrifugation rates, and instrument settings. Precautions were taken to avoid contamination with aluminum and iron and to prevent co-extraction of iron and aluminum not chelated to DFO. The addition calibration technique was used for standardization. Mean +/- SD recoveries of AIO and FO after extraction were respectively 98.3% +/- 3.7% and 101% +/- 3.7%. Sensitivity was 45 pg/0.0044 A.s for aluminum and 10 pg/0.0044 A.s for iron. Detection limits were 0.22 mumol/L for AIO, 0.06 mumol/L for DFO and FO.

Low percentage of aluminoxamine and ferrioxamine in uremic serum after desferrioxamine administration.

HPLC was used to study the effectiveness of two different desferrioxamine (DFO) administration strategies (15 mg/kg DFO, 1 h or 44 h before dialysis) on generation of aluminoxamine and ferrioxamine in five hemodialysis patients. The percentage of ultrafilterable aluminum and iron in these patients was also investigated by electrothermal atomic absorption spectrometry. The administration of DFO ...

Desferrioxamine provocative test: methodology for estimating iron and total iron binding capacity.

To the Editor: In our report entitled “Treatment of Cooley’s Anemia” (Blood 76:435, 1990), we described a desferrioxamine provocation test in which we measure serum iron and iron binding capacity following an infusion of the chelator. In the report, we failed to state clearly that we do not use standard colorimetric tests to make these measurements. The mild reducing agents used in most colorim...

Indirect Determination of Ascorbic Acid by Atomic Absorption Spectroscopy

A new and simple method followed by atomic absorption spectrometry has been developedfor indirect determination of ascorbic acid (AA). The proposed method was based on the oxidationof AA to dehydroascorbic acid with Cu2+cation(200mgL-1)in an ammonium thiocyanatesolution atpH2.4. AA reduces Cu2+to Cu+followed by the precipitation of Cu+ with SCN-. Then, the excess ofCu2+in the solution was measu...

Spectrophotometric determination of Dalacin phosphate from formulations using reduce atomic absorption of calcium ion by phosphate

A simple, sensitive and accurate method for spectrophotometric determination of Dalacinphosphatehas been established. The method is based on the reduction of atomic absorptioncalcium ion by phosphate. Because, there is a phosphate mole in every Dalacin mole structureand with measurement of phosphate, Dalacin is beyond measurement too. Therefore, the reactionbetween acidic hydrolysis products of...

Chelation of transferrin iron by desferrioxamine in K562 cells. The partition of iron between ferrioxamine and ferritin.

In this study we have determined whether desferrioxamine can chelate iron delivered to human leukaemic cells by the transferrin endocytic cycle. The cellular uptake of desferrioxamine was investigated by an indirect method in which the conversion of repeated pulses of [59Fe]transferrin to [59Fe]ferrioxamine was determined at two concentrations of the drug. Maximum generation of [59Fe]ferrioxami...
